Cytokine-induced reprogramming of human macrophages toward Alzheimer’s disease-relevant phenotypes

Brain-resident microglia and peripheral macrophages play key roles in neurodegenerative diseases such as Alzheimer’s disease (AD).
The researchers describe a cytokine-induced model of disease-associated microglia/lipid-associated macrophages (DLAMs).
Through transcriptomic and functional profiling of cytokine mix-treated THP-1 macrophages, they reveal a transition to the DLAM state, modulation of AD risk genes, and alterations in metabolism, lysosomal activity, and lipid homeostasis, providing insights into AD-related mechanisms.
