How Tau and α-Synuclein undermine synaptic health

Early defects in Alzheimer’s disease (AD) and Parkinson’s disease (PD), include Synaptic dysfunction.
Recent evidence suggests that Tau and α-Synuclein (α-Syn) may drive presynaptic pathology even before their aggregation.
In this review the researchers discuss the Tau and α-Syn mediated molecular defects at the presynaptic dysfunction.
Both Tau and α-Syn under normal conditions play important role in brain physiology but during disease, they acquire abnormal functions due to aberrant interactions and mislocalization.
The authors also highlight promising therapeutic targets aimed at maintaining synaptic function in both tauopathies and synucleinopathies.
