Lineage-tracing techniques have greatly advanced our understanding of pancreatic cell ontogeny and regenerative capacity, particularly in the context of βcells, which is paramount for developing therapeutic strategies for pancreatic diseases. 

Dual recombinase-based genetic lineage-tracing strategies provide enhanced precision and specificity in tracing cell fate, resolving controversies that have arisen with conventional lineage-tracing methods.

Three primary endogenous regenerative pathways of β-cells have been identified: self-duplication of preexisting β-cells, differentiation of endogenous adult stem cells, and reprogramming of non-β-cells into β-cells.

Lineage tracing of exocrine cells offers significant potential for advancing our understanding of the pathogenesis of pancreatitis and related diseases, paving the way for the development of new preventive and therapeutic strategies.
