Maintaining epidermal Langerhans cells
Autophagy is required for removal of harmful cytoplasmic components and mobilizes energy reserves in long-lived and self-renewing cells.
The researchers show that abrogation of autophagy in langerhans cells (LCs) leads to lipid accumulation & ferroptosis-mediated depletion, demonstrating that this pathway is necessary for LC maintenance in the epidermis.
Mechanistically, autophagic ATG5-gene deficient LCs showed metabolic dysregulation and accumulation of neutral lipids leading to the inability to process lipids and ferroptosis. LCs also showed upregulation of proinflammatory transcripts and showed decreased expression of neuronal interaction receptors.
