Portal vein-enriched metabolites in insulin resistance

Gut microbiome is implicated in diet and obesity mediated insulin resistance and type 2 diabetes.
To explore the role of gut-derived metabolites in this process, the researchers assessed portal/peripheral blood metabolites in mice with different risks of obesity/diabetes,
They show that circulating metabolites are differentially enriched in portal versus peripheral blood and the enrichment patterns are influenced by diet, host genetics, and the gut microbiome.
These metabolites, especially mesaconate/itaconate/ citraconate isomers, can directly influence hepatic insulin signaling and gene expression, highlighting an important role in obesity-linked insulin resistance.
The authors show that in humans, citraconate levels are inversely correlated with plasma glucose.
