Protein l-lactylation in metabolic regulation and cell signalling

Previously thought as a waste product, lactate act as a key metabolic intermediate and plays a crucial role in post-translational modification regulating cellular signaling.
The authors in this Perspective, discuss the enzymatic and metabolic mechanisms underlying protein lactylation and its impacts on both histone and non-histone proteins in the contexts of physiology and diseases.
They also discuss how protein lactylation plays a role in cellular functions such as cell-fate determination, development, cardiovascular diseases, cancer and autoimmune disorders.
The authors believe that protein lactylation could work as a potential therapeutic target in various diseases as it integrates metabolic and signalling pathways.
