Method for rapid vascular organoid development

For studying vascular development, disease, and regenerative medicine, development of vascular organoids (VOs) that controls both endothelial and mural compartments independently is very important.

The researchers in this study developed a rapid method to generate vascular organoids from human iPSCs by orthogonally activating the transcription factors ETV2 and NKX3.1.

The method produced functional 3D VOs in 5 days without ECM embedding but matured further upon ECM exposure, forming larger, structured vessels.

This strategy enabled controlled co-differentiation of vascular cells and supported in vivo engraftment, offering a versatile platform for vascular biology, tissue engineering, and regenerative medicine.
