How β2-adrenergic receptors regulate AMPA receptor trafficking and synaptic plasticity

Molecular mechanisms underlying norepinephrine (NE) mediated adrenergic receptor (AR) activation in attention is not well understood.

Surface expression of AMPA receptors (AMPARs) requires its complex formation with β2ARs, Gs, adenylyl cyclase, and protein kinase A (PKA) to augment AMPAR phosphorylation.

The researchers show that organic cation transporter 3 (OCT3) and plasma membrane monoamine transporter (PMAT) transport NE into neurons to stimulate β2ARs that are in their interior.

The ensuing phosphorylation of GluA1 on S845 by PKA drives AMPARs to the surface and postsynaptic sites during PKA dependent forms of LTP.
