What is Generalized pustular psoriasis?

Generalized pustular psoriasis (GPP) is a chronic, debilitating, and life-threatening skin condition posing unique clinical challenges due to a lack of therapeutic options to reliably achieve rapid and durable remission. 

The interplay between keratinocytes, neutrophils, T lymphocytes, and various cytokines (including IL-36 and IL-17) contributes to an autoinflammatory loop that distinguishes GPP from other types of psoriasis.

Several mutations along the IL-36 axis and its downstream pathways have been identified in individuals affected by GPP, resulting in neutrophil activation and dysregulated keratinocyte growth.

Recent advances in the research field of GPP include targeted therapeutics such as IL-36 blockade. The development of these candidate therapeutics is supported by genetic and immunological evidence. This is promising for carrying forward ongoing translational efforts to improve disease management and quality of care.
