While metabolic dysregulation is a known hallmark of malignancy, this research charts the exact molecular cascade linking dietary or circulating Branched-chain amino acid (BCAA)—specifically leucine—to lung tumorigenesis.

In Kras-mutant mice fed a high-BCAA diet showed aggressively accelerated lung tumor progression compared to controls. Leucine undergoes catabolism mediated by the enzyme AUH (RNA-binding protein/enoyl-CoA hydratase). This specific breakdown pathway generates an abundance of acetyl-CoA, a vital metabolic building block.

Instead of being used purely for energy, this acetyl-CoA fuels de novo cholesterol synthesis inside the cell. The newly synthesized cholesterol integrates into the cell membrane, promoting the formation of specialized microdomains called lipid rafts.

This structural change drives the redistribution and hyper-activation of the Epidermal Growth Factor Receptor (EGFR), a notorious driver of lung cancer growth.
