Immune–tumor synapse characterization for next-generation immunotherapies

The immunological synapse formed between cytotoxic CD8+ T lymphocytes and tumor cells constitutes the key site of action for immunotherapies.

Detailed characterization of the immunological synapse formed between cytotoxic CD8+ T lymphocytes and tumor cells has been challenging due to its dynamic nature.

Recently developed proteomics, transcriptomics, high-throughput imaging, and machine learning technologies have enabled a comprehensive analysis of the cytotoxic CD8+ T lymphocyte– tumor immunological synapse.

Integration of next-generation multiomics profiling, high-throughput imaging, and machine learning technologies is key for data-driven design of future immunotherapies that can overcome resistance and therefore improve patient response rates.
