SOX17 initiates an immune evasion program in colorectal adenomas and cancers by generating LGR5– cells and downregulating the interferon-gamma signaling pathway. 

Cell plasticity between LGR5+ and LGR5– cells is observed in both the normal intestine and CRCs.

Recent studies highlight the critical role of LGR5– cells in CRC liver metastasis.

Niche cells, including fibroblasts and lymphatic endothelial cells, are critical in shaping the tumor microenvironment and modulating tumor immunity.
