Protons inside the lysosomes makes them acidic but the source of protons is not clear.

The researchers introduce two molecular probes, AN-PZ for lysosomal digestion and PP-1 for mitochondrial depolarization, uncovering the proton transfer from mitochondria lysosomes through the membrane contact sites for lysosomal acidification.

The also found that bringing mitochondria and lysosomes physically into close proximity can increase lysosome acidification to enhance content digestion under disease conditions.
