Mechanisms regulating synaptogenesis are nit well understood.
The researchers performed image-based pooled CRISPR screens to elucidate mechanisms driving synapse formation.
This work established a scalable approach to map functional interactions guiding the activity of transsynaptic adhesion molecules and identified modulators of a synaptic organizer, neuroligin-1, implicating cell adhesion, cytoskeletal, and signaling processes in synaptogenesis.
The researchers show that the phosphatase PTEN and the dystrophin-associated glycoprotein DAG1 promote neuroligin’s roles in inducing presynaptic assembly, with DAG1 selectively regulating inhibitory synapses.
