Regulation of ferroptosis!

Ferroptosis is an iron-dependent form of programmed cell death that is driven by the accumulation of toxic lipid hydroperoxides. 

This study uncovers regulation of ferroptosis by the citrate transporter SLC25A1 and lyase ACLY, which cooperate to maintain stability of oxidoreductase FSP1 (AIFM2).

The researchers demonstrate that SLC25A1-ACLY axis sustains FSP1 acetylation, preventing it from proteasomal degradation and thereby reducing ferroptosis sensitivity.

They also show that FSP1 acetylation is dynamically controlled by the acetyltransferase KAT2B and the deacetylase HDAC3.

Pharmacological inhibition of SLC25A1 and ACLY significantly increases cancer cell vulnerability to ferroptosis in vitro and in vivo.
