The mechanism of acetaminophen action!

The mechanism of action of acetaminophen (paracetamol), a common analgesic, is not yet clear.
The study shows that acetaminophen counterintuitively inhibits diacylglycerol lipase α  (DAGLα), but not DAGLβ, reducing the production of the endocannabinoid 2-arachidonoyl glycerol.
This suggests a novel pain-relieving mechanism through cannabinoid 1 receptors.
These findings offer new insights into acetaminophen’s action, challenging traditional models of its analgesic effects.
