How high fructose promotes MYCN-amplified neuroblastoma progression

The researchers in this study evaluated fructose metabolite, acetate, on the transcriptional regulation of an oncogene, MYCN, gene expression in the neuroblastoma progression.

The researchers showed the role of acyl-coenzyme A (acyl-CoA) synthetase short-chain family member 2 (ACSS2)- mediated biosynthesis of acetyl-CoA in promoting MYCN-amplified neuroblastoma.

Mechanistically, ACSS2 orchestrates MYCN transcription by escalating acetyl-CoA levels and histone acetylation,  and  ACSS2 inhibitor blocked neuroblastoma progression. 

This study thus has opened the possibility of developing alternative strategies to suppress MYCN expression and halt tumor growth through interfering with metabolite-mediated regulation.
