Cell death or inflammation decision maker identified!
Activation of the Fas receptor can be prodeath or pro-inflammatory, but little is known about the switch between outcomes.
Cell death receptor Fas in addition to its pro death signals also sends out pro-inflammatory but how this switch happens is not clearly understood.
The researchers demonstrate that TLR adaptor CD14 can mediate this switch between death and inflammation in myeloid cells. The authors also show that CD14 acts as a crucial chaperone of Fas receptor internalization in macrophages and neutrophils, resulting in Cd14−/− myeloid cells that are protected from FasL-induced apoptosis, activate nuclear factor κB (NF-κB), and release cytokines in response.
The authors demonstrate that CD14 is also required for Fas to signal through the adaptor TRIF (TIR-domain-containing adapter-inducing interferon-β) and induce a pro-death complex.
