Principles of In vivo CAR T cell engineering
In vivo chimeric antigen receptor (CAR) T cells are a next-generation immunotherapy strategy designed to overcome the logistical, manufacturing, and accessibility limitations inherent to conventional ex vivo approaches.
A growing body of preclinical evidence demonstrates that lentiviral, lipid nanoparticle, and polymeric vectors can successfully generate functional CAR T cells in vivo and achieve durable disease control in preclinical models.
Both viral and nonviral delivery platforms have now entered clinical testing across diverse oncologic and autoimmune indications, with some early signs of efficacy reported.
Additional studies will be critical to determine long-term safety and durability as well as to establish rational design principles required for next- -generation in vivo CAR T cell products.
