Numerous distinct circular RNAs are generated largely via back-splicing in mammals, affecting various molecular, cellular, and physiological processes. 

While circular RNAs are inherently stable owing to their covalently closed topology lacking free 5′- and 3′-ends, they are targeted by various endoribonucleolytic pathways for degradation.

Beyond the regulation of their stability, circular RNAs intricately crosstalk with other RNA species through various direct and indirect pathways to control target molecules.

Given their high intrinsic stability and the diverse molecular mechanisms that regulate target RNA stability, such as microRNA sponging and circular RNA-induced nonsense-mediated mRNA decay, circular RNAs are emerging as an excellent and versatile platform for innovative RNA therapeutics.
