Small molecule transporter engineering and characterization
Membrane transporters serve fundamental roles in metabolite exchange. Engineering transporters can optimize precursor uptake and metabolite traffic, benefiting cell factory development. However, limited knowledge of transporter systems hampers efficient engineering, making transporter engineering challenging.
High-throughput transporter library screening and multi-omics analysis accelerate the identification of new transporter engineering targets and transporter functions. Furthermore, the evolutionary engineering of transporter proteins allows the generation of efficient transporter mutants.
Transporter characterization harnessing in vivo and in vitro systems gives rational insights into transporters’ substrate range and kinetics. Recent efforts have aimed at speeding up and simplifying the technologies for generating high quality data that guide rational transporter engineering.
