cGAS-STING signaling in neuroinflammation

The authors sin this review discuss the molecular mechanisms underlying the canonical innate immune responses that are triggered by cyclic GMP-AMP synthase–stimulator of interferon genes (cGAS-STING) pathway leading to the production of type I IFNs and pro-inflammatory cytokines.

They also discuss noncanonical roles of cGAS in chromatin organization and RNA metabolism.

cGAS-STING signaling also plays key roles in brain function, including glial state regulation, neuronal homeostasis, blood-brain barrier integrity, and peripheral immune surveillance, and neuroinflammation and neuropathology in neurodegenerative diseases.

The researchers also discuss the role of cGAS and STING inhibitors in treating the brain disorders.
