Protein homeostasis in aging neuron
Protein homeostasis, termed proteostasis, is essential for neuronal health, declines with age, contributing to neurodegenerative disease.
In this review, the authors evaluated the mechanisms of maintaining proteostasis network, how this is disrupted in ageing neurons, and its impact on neuronal function.
They also discussed unique challenges posed by neuronal compartments with distinct functions, as well as neurons’ high energy demands, and post-mitotic status and detail how proteostasis mechanisms differ across neuronal compartments and neural subtypes, and how these differences influence susceptibility to stress and disease.
The authors also explore how these differences shape selective vulnerability in neurodegenerative diseases and how compartment- and cell-type-specific approaches should be pursued towards therapeutic goals in treating aging brain.
