A human tumor-immune glioblastoma organoid model
There is a lack of human-specific, immunocompetent glioblastoma model that reflect the human tumor microenvironment.
The researchers introduce immune-human organoid tumor transplantation (iHOTT) model, an autologous organoid co-culture system using patient-derived tumor cells and matched peripheral blood mononuclear cells to preserves human tumor-immune interactions and T cell clonal responses.
iHOTT mirrors cell-type shifts and cell-cell interactions observed in patients to PD-1 blockade (using pembrolizumab) and TCR profiling in iHOTT reveals pembrolizumab-driven expansion of CD4 T cell clonotypes,
iHOTT thus provides a platform for evaluating personalized immunotherapy strategies in glioblastoma.
