Macrophages are implicated in the pathogenesis of obesity, yet the underlying mechanisms remain incompletely understood. 

The authors show that prostaglandin E2 receptor subtype 3 (EP3) in macrophages is shown to be a critical regulator of diet-induced adipogenesis.

They demonstrate that EP3 expression is decreased in adipose tissue macrophages from patients with obesity and high-fat diet (HFD)-fed mice.

Macrophage-specific deletion of EP3 promotes fat cell differentiation and exacerbates HFD-induced obesity.

The authors reveal that EP3 depletion decreases expression of matricellular protein Sparc in macrophages through promotion of Dnmt1/3a-mediated DNA methylation.

Pharmacological activation of EP3 receptor alleviates HFD-induced obesity in mice.
