Prime editors for mammalian genome editing

The newly developed prime editing system, called pvPE, which uses a reverse transcriptase derived from porcine endogenous retrovirus, outperforms other reported prime editing systems in both editing efficiency and precision. 

Multiple optimization strategies boosted editing efficiency by 24.38–101.69-fold compared with the original pvPE across diverse mammalian cell lines.

A small molecule, nocodazole, significantly enhanced pvPE editing efficiency by modulating the DNA repair pathway.

Using the pvPE + Nocodazole system and somatic cell nuclear transfer, we generated triple gene-edited pigs that could serve as a valuable model for research into Alzheimer’s disease in humans.
